Expression and Significance of MMP-7 and PTEN Protein in Primary Human Hepatocellular Carcinoma

  • 摘要: 目的 探讨基质金属蛋白酶7(matrilysin,MMP-7) 和PTEN在原发性肝细胞性肝癌(HCC)中的表达状况,并探讨他们之间的关系及其临床意义。 方法 应用免疫组织化学SP法检测原发性肝细胞性肝癌150例、肝硬化50例、肝炎50例、正常肝组织30例中MMP-7、PTEN的表达。 结果 MMP-7,PTEN主要表达于细胞质,MMP-7和PTEN在原发性肝细胞性肝癌、肝硬化、肝炎和正常肝组织的表达差异有统计学意义,且MMP-7在原发性肝细胞性肝癌组织中的表达明显高于在正常肝组织中的表达(χ2=20.17,P< 0.05),而PTEN的阳性表达率明显弱于正常肝组织(χ2 =16.26,P<0.05)。MMP-7,PTEN关系密切,呈负相关。 结论 PTEN、MMP-7的表达一定程度上反映HCC侵袭性强弱,PTEN缺失可引起MMP-7表达增加,在HCC侵袭、转移中发挥作用。

     

    Abstract: Objective To explore the expression and clinical significance of MMP-7(matrilysin) and PTEN protein in primary human hepatocellular carcinoma. Methods The expressions of MMP-7(matrilysin) and PTEN protein in 150 HCC specimens, 50 cirrhosis specimens, 50 hepatitis specimens and 30 normal specimens were detected by immunohistochemistry method. Results The rate of expressions of MMP-7 protein in cancer tissues, cirrhosis tissues, hepatitis tissues and normal tissues were respectively 47.33%(71/150),26.00%(13/50),18.00%(9/50),3.00%(1/30)and significantly different (χ2=31.83, P <0.05). The expression of MMP-7protein in cancer tissues cirrhosis tissues was higher than that in normal tissues (χ2=20.17, P<0.05). The rate of expressions of PTEN protein in normal tissues, cirrhosis tissues, hepatitis tissues and cancer tissues were respectively 100%(30/0),66.00%(33/50),82.00%(41/50),62.67%(94/150) and significantly different(χ2=20.99, P<0.05).The expression of PTEN protein in cancer tissues was weaker than in normal tissues(χ2=16.26,P<0.05). Conclusion The expression of MMP-7 is closely associated with the expression of PTEN in HCC and they are negatively corre-lated; The abnormal expression of MMP-7 and PTEN are closely associated with the development of HCC.They may play important roles in development of HCC.
