Expression of Dickkopf1 in Two Human Glioma Cell Lines

  • 摘要: 目的 检测Dickkopf1(DKK1)在人胶质瘤细胞株MGR3和UWR7中的表达情况。 方法 采用酶联免疫吸附实验,逆转录多聚酶链反应等方法对DKK1在人胶质瘤细胞株MGR3和UWR7中表达水平进行检测。 结果 酶联免疫吸附实验提示DKK1在MGR3和UWR7中均呈高浓度;逆转录多聚酶链反应提示DKK1基因在胶质瘤细胞株MGR3和UWR7中高表达,但在正常脑组织中表达不明显。 结论 DKK1在人胶质瘤细胞株MGR3和UWR7中高表达,提示DKK1可能在胶质瘤中的发生发展中起重要作用。

     

    Abstract: Objective To detect the expression of dickkopf1 (DKK1) in human glioma cell lines MGR3 and UWR7. Methods The expression of DKK1 in human glioma cell lines MGR3 and UWR7 was detected by enzyme-linked immunosorbant assay (ELISA) and reverse transcriptase-polymerase chain reaction (RT-PCR). Results ELISA results demonstrated that the expression of DKK1 was both up-regulated in MGR3 and UWR7.RT-PCR results showed that DKK1 transcription level was high in MGR3 and UWR7, but was not detectable in normal brain tissues. Conclusion DKK1 is over-expressed in glioma cell lines MGR3 and UWR7,which may play an important role in the initiation and development of glioma.
