Screening of Upstream Regulators of Proto-Dbl Oncoprotein by Yeast Two-hybrid System

  • 摘要: 目的:探讨VL domain对Dbl癌基因活性的调控机制。方法:本文通过酵母双杂交的方法,以proto-Dbl的VL domain为诱饵蛋白对人胎脑文库进行筛选。结果:研究共获得7个能与VL domain相互作用的蛋白。结论:Proto-Dbl VL domain相互作用蛋白的发现将为揭示proto-Dbl的功能奠定基础。

     

    Abstract: Objective:The VL domain truncate mutation could directly cause the activation of proto-Dbl and the malignant transformation. Methods:In order to clarify the activation mechanism of proto-Dbl, yeast two-hybrid system was used to screen the proteins interacted with the VL domain in the human fetal brain cDNA library taking VL domain as the bait. Results:7 positive clones which interact with proto-Dbl-VL domain were successfully harvested. Conclusion:This discovery lays the foundation for uncovering of the regulatory mechanism of proto-Dbl.
