转染KISS-1对裸鼠食管鳞癌移植瘤生长的 影响

Effects of KISS-1 Transfection on Tumor Growth of Nude Mice

  • 摘要: 目的 探讨KISS-1基因对裸鼠食管鳞癌移植瘤生长的影响。方法 通过脂质体介导将pcDNA3.1-KISS-1转染人食管鳞癌细胞EC-1,用Western blot法检测转染前、后KISS-1蛋白的表达。将单纯EC-1细胞、稳定转染空质粒的EC-1细胞、稳定转染pcDNA3.1-KISS-1的EC-1分别接种于BALB/c-nude裸鼠,构建裸鼠食管鳞癌移植瘤模型,观察肿瘤生长情况,测量肿瘤体积和瘤质量并绘制肿瘤生长曲线;免疫组织化学和半定量RT-PCR技术检测肿瘤组织中KISS-1蛋白和mRNA的表达。结果 KISS-1基因转入食管鳞癌EC-1细胞,KISS-1蛋白的表达(1.143±0.218)较空质粒转染组(0.745±0.130)和对照组(0.855±0.184)明显增加(P<0.05),转染KISS-1基因组裸鼠较空质粒转染组和对照组肿瘤生长速度慢,体积(949.42±102.26)mm3与空质粒转染组(1339.82±72.23)mm3和对照组(1384.04±97.07)mm3相比明显减小(P<0.05),瘤体重量(0.498±0.654)g与空质粒转染组(0.638±0.066)g和对照组(0.676±0.108)g相比明显减轻(P<0.05)。结论 通过基因转染的方法能表达有活性的KISS-1,并能抑制裸鼠食管癌移植瘤的生长。

     

    Abstract: Objective To investigate the effects of KISS-1 transfection on the tumor growth of nude mice. Methods Esophaged squamous carcinoma cell EC-1 was transfected with KISS-1 by liposome, and KISS-1 protein in transfected and nontransfected cells was detected by Western blot. Nude mice were transplanted subcutaneously with EC-1 cells, EC-1 stably transfected with empty vectors, EC-1 cells stably transfected with KISS-1, respectively. The tumor volume and weight were measured. Immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR) were used to detect KISS-1 protein and mRNA level in tumor tissue. Results Expression of KISS-1 protein in pcDNA3.1-KISS-1 transfected cells (1.143±0.218) was significantly higher than that in pcDNA3.1 transfected cells (0.745±0.130) and control cells (0.855±0.184)(P<0.05). In vivo study showed the tumor growth speed of nude mice in pcDNA3.1-KISS-1 transfected group was significantly slower than that in pcDNA3.1 transfected group and control group. The tumor volume in pcDNA3.1-KISS-1 transfected group (949.42±102.26)mm3 was significantly smaller than that in pcDNA3.1 transfected group (1339.82±72.23)mm3 and control group (1384.04±97.07)mm3(P<0.05). The tumor weight in pcDNA3.1-KISS-1 transfected group (0.498±0.654)g was significantly lighter than that in pcDNA3.1 transfected group (0.638±0.066)g and control group (0.676±0.108)g(P<0.05). Conclusion KISS-1 gene can effectually express after gene transfection and may obviously inhibit the growth of the xenograft tumor of nude mice.
