枸杞多糖增强人树突状细胞抗肿瘤的机制

Anti-tumor Mechanism of Human Dendrtic Cells by Lycium Bararum Polysaccharides

  • 摘要: 目的 观察枸杞多糖对树突状细胞抗肿瘤的增强作用。 方法 分离人外周血单核细胞,体外培养其成为未成熟树突状细胞和成熟树突状细胞。用冻融法制备肝癌细胞的全抗原并致敏未成熟树突状细胞;通过混合淋巴细胞实验,获取成熟树突状细胞激活的特异性效应T细胞。用MTT法测定效应T细胞对肝癌细胞的杀伤率。观察枸杞多糖在树突状细胞的成熟及T细胞的激活、增殖和杀瘤过程中的作用。 结果 枸杞多糖能促进树突状细胞的成熟, 转染了人肝癌细胞的全抗原的mDC可特异地激活T细胞为效应T细胞。效应T细胞可特异性地杀伤人肝癌细胞,而对不相关的MG-63细胞无特异性的杀伤作用。单独对T细胞无明显的增殖能力,但并且能增强效应T细胞的增殖能力和杀瘤活性。 结论 枸杞多糖能促进树突状细胞成熟和增强效应T细胞的增殖和杀伤肿瘤的活性。

     

    Abstract: Objective To investigate the enhanced anti-tumor effect of the dendritic cells by lycium bararum polysaccharides. Methods Monocytes obtained form human peripheral blood were induced into immature DCs and mature DCs in vitro. The whole antigen of hepatocarcinoma cells was extracted by freeze thawing method and used to sensitize immature DCs; Effective T cells were harvested by mixed lymphology reaction, and the specificity that effective T cells killed target hepatocarcinoma cells was examined by the MTT method. The effects of lycium bararum polysaccharides on DCs' maturity,T cell activation,proliferation and anti-tumor were studied. Results Lycium bararum can promote the maturity of DCs, DCs with whole antigen of hepatocarcinoma cells can activate specifically T cells into effective T cells which can kill hepato carcinoma cells specifically while not MG-63 cells. polysaccharides alone had no significant effect on the proliferation of T cells but they could promote the maturity of DCs and reinforce proliferation and anti-tumor effects of effective T cells. Conclusion Lycium bararum polysaccharides can promote the maturity of DCs and reinforce proliferation and anti-tumor effects of effective T cells.
