硫化氢对人肝癌细胞增殖和凋亡的影响

Effects of Hydrogen Sulfide,A New Gaseous Signal Molecule,on Proliferation and Metabolism of HepG2

  • 摘要: 目的 观察新型气体信号分子硫化氢对肝肿瘤HepG2细胞增殖和凋亡的影响,并探讨其作用机制。 方法 将不同浓度的硫氢化钠体外作用于体外培养的人肝癌细胞HepG2后,显微镜观察HepG2细胞形态改变,四甲基偶氮唑蓝(MTT)比色法检测肝癌细胞增殖,流式细胞仪检测肝癌细胞周期,DNA片段化检测细胞凋亡及不同浓度蛋白激酶抑制剂下硫氢化钠对细胞的增殖作用。 结果 在5 mmol/L~10-4 mmol/L不同浓度硫氢化钠作用下,人肝癌细胞均增殖,其中以1 mmol/L浓度的硫氢化钠效果最明显;细胞周期中以G1为主。在1 mmol/L NaHS的培养下,受不同浓度的蛋白激酶抑制剂(Tyrphostin 47)预处理后,随着Tyrphostin 47浓度增加,细胞的生长愈受到抑制。 结论 硫化氢能诱导培养的肝肿瘤细胞生长,其机制可能是通过激活蛋白激酶,诱导细胞周期增殖,其中1 mmol/L浓度促细胞增殖作用最明显。

     

    Abstract: Objective To investigate the effects of hydrogen sulfide(H2S)on the proliferation and metabolism of human hepatocelluar carcinoma HepG2 cells. Methods HepG2 cells was treated with NaHS at different concentration.The cellular ultrastructure was observed under the inverted phase contrast microscope. Multiparameter such as proliferation index (PI), cell cycle,cell apoptosis and growth inhibition rate was detected by typan blue staining, MTT and flow cytometry (FCM). Results Various concentrations from 5 mmol/L to 10-4mmol/L of NaHS promoted HCC cell growth and seldom induced cell apoptosis. G1 stage was increased, while S stage was reduced. The effect of NaHS in combination was significant (P<0.05). The most effective concentration of NaHS was 1 mmol/L. Under the concentration of 1mmol/L NaHS, the growth of HepG2 cell was restrained with the increase in the concentration of Tyrphostin 47. Conclusion H2S may induce the growth of HCC cell, and its mechanism was associated with the activation of mitogen-activated protein kinase.The most effective concentration of NaHS that promote cell proliferation was 1 mmol/L.
