Effect of SerpinH1 on Proliferation and Growth of Murine Tumor

  • 摘要: 目的 研究SerpinH1基因对肿瘤细胞增殖的影响。 方法 克隆SerpinH1基因,建立相关稳定转染细胞系,分为5组,分别为空白对照组,pcDNA 3.1/V5-SerpinH1实验组,pcDNA 3.1/V5对照组,pNKRY-SerpinH1-siRNA实验组,pNKRY-GFP对照组。应用BrdU渗入法,流式细胞术检测,WST-1方法分别测定细胞增殖情况,并建立肿瘤细胞模型,用体内试验验证SerpinH1对肿瘤细胞生长情况的影响。 结果 过表达基因的pcDNA 3.1/V5-SerpinH1组肿瘤生长有抑制作用,而用siRNA干扰技术沉默基因表达的pNKRY-SerpinH1-siRNA组肿瘤生长有促进作用。 结论 SerpinH1基因能够抑制肿瘤细胞生长,提示SerpinH1具有抑制肿瘤的功能。

     

    Abstract: Objective To investigate the effects of SerpinH1 on the proliferation of murine tumor. Methods We first cloned SerpinH1 and set up SerpinH1 targeted siRNA structure, and then investigated the effects of SerpinH1 on the tumor cell proliferation and growth. The inhibition of cell proliferation and growth was demonstrated by both WST-1 cell proliferation assay and 5′-bromo-2′-deoxy-uridine incorporation assay with flow cytometry (FACS). Results The expression of SerpinH1 could inhibit the proliferation of the monocyte tumor RAW264.7 cells and melanoma B16 cells. Meanwhile silencing SerpinH1 could promote the proliferation of both RAW264.7 and B16 cells. Conclusion The results indicated that SerpinH1 could inhibit the proliferation and growth of both RAW264.7 and B16 tumor cells.
