胶质瘤钴60照射后脑水肿与erb-B1蛋白表达的关系

Relationship between Cerebral Edema and erb-B1 Protein Expression in Glioma Rats Treated with 60Co Irradiation

  • 摘要: 目的 研究脑胶质瘤大鼠钴60(60Co)照射后,出现放射性脑水肿的比例、程度及分布与erb-B1基因表达变化的关系。 方法 建立大鼠脑胶质瘤模型,将荷瘤大鼠随机分成对照组、实验组和空白组。15天后,对照组和实验组给予60Co 照射,实验组60Co 照射前后给予ZD1839(erb-B1抑制剂)治疗,空白组不给照射和药物治疗。免疫组织化学染色及计算图像分析检测大鼠脑水肿情况和erb-B1蛋白表达水平。其中,空白组记为A组,实验组中无脑水肿大鼠记为B组,实验组中明显脑水肿大鼠记为C组,对照组中无脑水肿大鼠记为D组,对照组中明显脑水肿大鼠记为E组。 结果 胶质瘤荷瘤大鼠erb-B1蛋白表达水平:C组高于B组、E组高于D组,差异均有统计学意义(P<0.05);A组低于C组和E组,差异有统计学意义(P<0.05)。对照组脑水肿率明显高于实验组,差异有统计学意义(P<0.05)。 结论 胶质瘤60Co照射后,放射性脑水肿的发生与erb-B1蛋白表达水平有关。

     

    Abstract: Objective To observe the relationship between cerebral edema and erb-B1 protein expression in glioma rats treated with 60Co irradiation. Methods Glioma rats model was built. After 15 days the negative group was treated with 60Co,the experiment group was treated with 60Co and ZD1839. Immunohistochemical technique SP staining was used to check erb-B1 protein expression.With computer image analysis. Group A was the blank group, group B included the none edema rats in experiment group,group C included the serious edema rats in experiment group, group D had the none edema rats in control group, group E caontained the serious edema rats in control group. Results The levels of erb-B1 protein expression were significantly different between group C and B, group E and D,group A and C,group A and E (P<0.05). The serious edema percentages were also significantly different between the control group and the experiment group (P<0.05). Conclusion The mechanism of radioactive cerebral edema is tightly connected with erb-B1 protein expression after 60Co therapy.
