E2F-1和Rb基因在食管鳞状细胞癌中的 表达及临床意义

Expression and Clinical Significance of E2F-1 and Rb Genes in Esophageal Squamous cell Carcinomma

  • 摘要: 目的 研究E2F-1和Rb基因在食管鳞状细胞癌中的表达情况,探讨其在食管鳞状细胞癌发生发展中的规律与临床病理关系。方法 应用免疫组化方法检测60例食管鳞状细胞癌和50例正常食管组织中E2F-1和Rb基因的表达情况。结果 食管鳞状细胞癌中E2F-1的阳性表达率为73.3%,显著高于癌旁正常组织黏膜中的36.0%(P<0.01),E2F-1的表达与食管鳞状细胞癌的分期、分化及淋巴结转移有相关性(P<0.05)。Rb在食管鳞状细胞癌中的阴性表达率为30.0%,显著高于癌旁正常组织中的12.0%(P<0.05),Rb在食管鳞状细胞癌中的表达与分化及淋巴结转移有关(P<0.05),但尚不能认为与分期有关。结论 E2F-1和Rb在食管鳞状细胞癌中的表达呈负相关,即E2F-1过度表达而Rb表达有一定程度缺失,在正常食管组织中的表达亦有差异。

     

    Abstract: Objective To investigate the expressions of E2F-1 and Rb in the esophageal squamous cell carcinomma and approach the relationship between the genes and clinical pathology and the roles in the process of esophageal squamous cell carcinogenesis. Methods Immunohistochemistry was used to detect the expressions of E2F-1 and Rb in 60 cases of esophageal squamous cell carcinoma and 50 cases of normal esophageal tissue. Results The positive rate of E2F-1 expression was 73.3% in cancerous tissue, which was significantly higher than 36.0% in normal esophageal tissue(P<0.01). The expression of E2F-1 was closely associated with tumor stage, differentiation and lymph node invasion(P<0.05).The negative rate of Rb was 30.0%in cancerous tissue,which was significantly higher than 12.0% in normal esophageal tissue(P<0.05).The expression of Rb was closely associated with tumor differentiation and lymph node invasion(P<0.05),but not associated with tumor stage. The expressions of E2F-1 and Rb in esophageal squamous cell carcinoma was negatively correlated (r=0.395,P=0.02). Conclusion The overexpression of E2F-1 and decreased expression of Rb involve in the occurrence and development of esophageal carcinoma in some extent.
