Relationship between Expression of SDF-1 in Liver and Metastasis of Colorectal Carcinoma

  • 摘要: 目的 探讨SDF-1在人体不同正常组织中的表达差异以及这种差异的临床意义,探讨SDF-1在结直肠癌患者肝脏组织中的表达及其临床意义。方法 应用免疫组化方法对30例3种不同人体组织的SDF-1的表达进行检测、分析SDF-1在不同组织中的表达;应用免疫组化方法对67例结直肠癌中原发组织及其正常的肝脏组织和25例肝转移癌组织标本中的SDF-1、CXCR4的表达进行研究,观察SDF-1/CXCR4在结直肠癌肝转移中的作用。结果 SDF-1在皮肤组织中的阳性表达率为20%(6/30),在小肠黏膜中的阳性表达率为26.7% (8/30),在肝组织中的阳性表达率为83.3%。SDF-1在正常人体肝组织中的表达明显高于其他两种组织(P<0.05);SDF-1在结直肠癌患者的正常肝脏组织中的阳性率为80.9% (54/67),其表达与结直肠癌肝转移的发生率有关,表达高者,转移率高。结论 正常肝组织中CXCR4特异性配体SDF-1表达升高,而高表达SDF-1的肝组织可能成为表达CXCR4的结直肠癌转移靶点。SDF-1/CXCR4在结直肠癌肝特异性转移过程中起重要的作用。

     

    Abstract: Objective To study the expression of SDF-1 in different normal human tissues and its clinical significance; to indicate the expression of SDF-1 in normal hepatic tissue of colorectal carcinoma patients and its clinical significance. Methods Immunohistochemistry SP method was used to detect the expression of SDF-1 in three different kinds of human normal tissues of 30 cases, and the expression of SDF-1 and CXCR4 in 67 colorectal carcinoma tissue and their normal hepatic tissues tumor and 25 liver metastasitic tissue in colorectal carcinoma. The relationship between the expression of SDF-1 and metastasis of colorectal carcinoma was observed. Results The positive expression rates of SDF-1 was different in the skin tissue, the small intestinal mucosa and hepatic tissue, which was 20%,26.7% and 83.3% respectively; the positive expression rates of SDF-1 in the hepatic tissue was significantly higher than that in other tissues (P<0.05);the positive expression rates of SDF-1 in the normal liver tissue of colorectal carcinoma patients was 80.9% (54/67). The expression of SDF-1 in liver was correlated with the rate of liver metastatic in colorectal cancer. The higher expression of SDF-1, the more liver metastasis in colorectal cancer. Conclusion The hepatic tissue becomes metastatic aim of colorectal carcinoma because of its high expression of SDF-1. The high expressions of SDF-1/CXCR4 may play an important role in the process of colorectal carcinoma metastasis to liver.
