XIAP在宫颈鳞状细胞癌中表达及动脉介入化疗对其的影响

Effect of Intra-arterial Influsion Chemotherapy(IAIC) on Expression of X-linked Inhibitor of Apoptosis Protein (XIAP) in Squamous Cell Carcinoma of Cervix

  • 摘要: 目的 探讨X连锁凋亡抑制蛋白(XIAP)在宫颈鳞状细胞癌中表达的意义及动脉介入化疗对宫颈鳞状细胞癌(XIAP)表达的影响。 方法 选取50例宫颈鳞癌患者介入化疗前后宫颈组织,以30例正常宫颈组织为对照,采用免疫组化染色SP法检测XIAP表达水平的变化,采用DNA缺口原位末端标记法(TUNEL)检测细胞凋亡指数(AI)。 结果 XIAP在宫颈鳞癌细胞的表达明显高于正常宫颈组织(P<0.05),且在低分化癌组织中表达明显高于高、中分化癌组织,Ⅲ期鳞癌明显高于Ⅱ期(P<0.05),介入化疗前XIAP表达水平明显高于介入化疗后(P<0.05),而介入化疗前AI明显低于介入化疗后(P<0.05)。 结论 XIAP在宫颈鳞状细胞癌的发生发展中起着重要作用,动脉介入化疗可能通过有效下调XIAP表达,从而诱导宫颈鳞癌细胞凋亡。

     

    Abstract: Objective To investigate the expression of the X-linked inhibitor of apoptosis protein (XIAP) in squamous carcinoma of cervix and the effect of Intra-arterial influsion chemotherapy on XIAP. Methods Samples from 30 normal cervical tissues and 50 cervical squamous carcinoma tissues before and after Intra-arterial influsion chemotherapy were collected. The expression of XIAP was detected in these samples by SP immuno-histochemical technique. Apoptosis index(AI) was detected by DNA in situ terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL). Results The XIAP expression in squamous carcinoma of cervix was significantly higher than that in normal cervical tissue (P<0.05). Samples with well-differentiated tumor had lower XIAP expression than poorly differentiated one(P<0.05).It was also coincident increased with squarmous cell carcinoma staging or degree of malignancy. Patients after the treatment by IAIC had lower expression of XIAP than the one before IAIC treatment (P<0.05). Conclusion Abnormal expression of XIAP may contribute to the pathogenesis and development of the squamous carcinoma of cervix.It suggested that apoptosis induced by intra-arterial influsion chemotherapy may mediated by the downregulation of XIAP in cervical squamous cell carcinoma.
