Expression and Clinical Significance of Midkine in Thyroid Gland Carcinoma

  • 摘要: 目的 研究妊娠中期因子(MK)在甲状腺癌中的表达及其临床意义。方法 应用RT-PCR、原位杂交方法对50例人甲状腺癌组织、30例良性肿瘤组织、30例正常口腔组织进行了MKmRNA表达的研究。结果 RT-PCR分析显示MKmRNA在甲状腺癌组织表达量远高于良性肿瘤组织(P<0.01)。本实验中缩短型MK(tMK)仅表达于癌组织。 原位杂交结果显示MK的阳性产物位于癌细胞胞浆中;甲状腺癌组织中MK的表达率明显高于良性肿瘤组织(P<0.01);转移组MK的表达率明显高于未转移组(P<0.01)。分类及分期观察,MK表达量随着甲状腺癌分期级别的递增呈上升趋势。结论 MKmRNA在甲状腺癌组织中高表达,并与疾病的恶性度密切相关;tMK可以作为恶性肿瘤标记物;检测MK对临床疾病诊治方案的确定具有参考价值。

     

    Abstract: Objective To investigate the expression and clinical significance of midkine mRNA in thyroid gland carcinoma. Methods RT-PCR and in situ hybridization techniques were employed to detect the expression of MK mRNA in 50 cases of thyroid gland carcinoma samples, 30 cases of benign tissue and 30 cases of normal tissue. Results MKmRNA was only detected in thyroid gland carcinoma but normal tissues by RT-PCR, it also showed that MK expression were completely different between cancer and benign tissues (P<0.01). In situ hybridization analysis demonstrated that tMK expression was mainly laid in the cytoplasm of cancer cells, which was significantly higher than the control groups (P<0.01). Furthermore, we have found that in thyroid gland carcinoma MK expression was coincidently increased with upgrade tumor classification and staging. Conclusion MK was highly expressed in thyroid gland carcinoma, and was positively correlated with the progression of malignancy disease. Our results showed that examination of MK could be a valuable marker for the clinical diagnostic purpose and treatment of thyroid gland carcinoma disease.
