骨桥蛋白在人骨肉瘤中的表达及其临床意义

Expression of Osteopontin mRNA and Its Clinical Significance in Osteosarcoma

  • 摘要: 目的 研究骨肉瘤中骨桥蛋白(OPN)mRNA的表达及其与临床病理特征之间的关系,阐明骨桥蛋白在骨肉瘤侵袭转移中的作用。方法 应用半定量RT-PCR方法。检测OPN mRNA在58例骨肉瘤组织和25例瘤旁相对正常组织中的相对表达量,分析OPN mRNA的相对表达量与骨肉瘤临床病理特征的关系。结果 58例骨肉瘤组织标本中,OPN mRNA高表达率为81.0%(47/58),而25例瘤旁相对正常组织标本中OPN mRNA高表达率仅16.0%(4/25)。OPN mRNA的表达与骨肉瘤的分化程度、侵袭范围、年龄以及GTM分期密切相关,而与患者性别、肿瘤部位、肿瘤直径无关。58例患者随访12~24个月,平均18个月。术后复发23例,死亡8例。其中OPN mRNA高表达组47例中复发20例,死亡7例;而低表达组11例中复发3例,死亡1例。结论 骨肉瘤中骨桥蛋白mRNA的表达明显增高。OPN mRNA的高表达反映了骨肉瘤病情的进展,并且与患者的预后有关,提示OPN可能是骨肉瘤的一种重要负性预后因子。骨桥蛋白在骨肉瘤远处转移过程中的作用有待进一步研究。

     

    Abstract: Objective  To investigate the expression of osteopontin mRNA and it s correlation with clinicopathologic features of osteosarcoma and elucidate its role in tumor invasion and distant metastasis. Methods  The expression of OPN mRNA was detected by semi-quantitive RT-PCR. The relationship between the relative content of OPN mRNA and clinicopathologic features of osteosarcoma was analyzed. Results  In 58 osteosarcoma tissue samples, the rate of OPN mRNA high-level expression was 81. 0 % (47/ 58) .However, the high-level expression rate in all 25 cases of relative normal osteosarcoma was 16. 0 % (4/25) . The expression of OPN mRNA was associated with the differentiation degree, scope of tumor invasion, years of age and GTM staging, but had no correlation with sex, tumor location and diameter. The 58 patients were followed up for 12~24 months (mean 18 months) . The OPN mRNA high-level expression group (47 cases) had recurrence in 20 patients and death in 7 patient s, whereas the low-level expression group (11 cases) had only 3 cases with recurrence and 1 case dead. Conclusion  OPN mRNA is overexpressed in osteosarcoma and it reflects the progression of diseases and association with poor prognosis of osteosarcoma. The role of OPN playing in the process of distant metastasis of osteosarcoma is required a further study.
