缺氧时缺氧诱导因子-1α在胃癌细胞的表达及其同细胞增殖凋亡的关系

Expression of HIF-1α and Its Relationship to Apoptosis and Proliferation in Gastric Cancer Cell During Hypoxia

  • 摘要: 目的研究缺氧诱导因子-1α(hypoxia-induciblefactor-1α,HIF-1α)在缺氧的胃癌细胞SGC-7901的表达及其同缺氧时Fas、PCNA关系,探讨HIF-1α同缺氧时胃癌细胞增殖凋亡的关系。方法产气袋法(GasPaK)制造缺氧细胞培养条件,细胞免疫化学观察SGC-7901细胞HIF-1α、PCNA及Fas的变化。结果缺氧时SGC-7901细胞HIF-1α表达明显上调,HIF-1α表达随缺氧时间延长而增加。缺氧时HIF-1α表达与Fas呈负相关(P<0.05),同PCNA表达无关。结论缺氧能使SGC-7901细胞HIF-1α表达明显上调,HIF-1α抑制缺氧诱导的细胞凋亡,同缺氧时胃癌细胞增殖无关。

     

    Abstract: Objective Investigated the expression of HIF-1α and its relationship to Fas and PCNA, and studied the HIF-1α and its relationship to apoptosis and proliferation in gastric cancer cell during hypoxia. Methods The environment of hypoxia was established by GasPak method. Cell-immunochemistry were used to observe the change of HIF-1α、PCNA and Fas.
