Expression of Fas/APO -1 in human esophageal cancerous cells transducted with Fas/APO-1 gene

  • 摘要: 目的 通过将Fas/APO 1基因转染食管癌细胞, 比较转导前后基因及蛋白的表达水平。方法 运用已构建的Fas/APO 1真核表达质粒, 用脂质体介导法将目的基因导入Eca 10 9细胞, 筛选克隆细胞以Southernblot, Northernblot, Westernblot法检测Fas/APO 1基因的表达。结果 转导株与非转导株均有Fas/APO 1cDNA的表达, 但转导株在基因及蛋白水平的表达均明显高于非转导株。结论 Fas/APO 1基因在食管癌细胞中处于低表达状态 ;通过基因转导能有效增强其表达。

     

    Abstract: Objective To compare the expressing level of Fas/APO -1 gene in gene transfected and not transfected esophageal cancer cells through transducting Fas/APO -1 gene into esophageal cancer cells. Methods The reconstructed plasmid expressing Fas/APO -1 gene with lipofectamine was transducted into esophageal cancer cell line Eca-109.
