钙通道阻滞剂对体外培养的脑膜瘤细胞信号转导及增殖的影响

Effects of Calcium Channel Antagonist on Signal Transduction and Proliferation of in Vitro Meningioma Cell

  • 摘要: 目的 观察维拉帕米对体外培养的脑膜瘤细胞信号转导及增殖的影响并探讨其作用机制。方法 采用荧光光度计检测脑膜瘤细胞内游离钙离子浓度(Ca2+i)水平,结合细胞计数方法测定细胞的增殖情况。结果 1μmol/L维拉帕米能明显抑制血清诱导的脑膜瘤细胞的增殖(P<0.01);细胞数为对照组的80%。10%的血清作用于脑膜瘤细胞以后,Ca2+i迅速升高到(435.3±34.9)μmol/L(P<0.01)。用维拉帕米预处理后,Ca2+i为221.1±26.1,升高辐度明显降低,(P<0.01)。结论 维拉帕米能抑制血清诱导的体外培养的脑膜瘤细胞增殖信号的转导,其作用机制与降低了Ca2+i增加有关。

     

    Abstract: Objective  This study aimed to assess the effects of verapamil on signal transduction and proliferation of in vitro meningioma and inquired into the possible mechanisms. Methods  Spect rofluorometer was used to measure the cytoplasmic free calcium; cell count was used to determine the proliferation of meningioma. Results  Verapamil in concent ration of 1μmol/ L l can block the proliferation of in vitro meningioma cell induced by FCS, the rate of cell number (vs. cont rol group) is 80 %, FCS induced a quick rise of int racellular free calcium ( P < 0. 01) ; Ca 2+ I was increased to 435. 3 ±34. 9μmol/ L, preincubation with verapamil could inhibit the rise of Ca2+ I, the Ca2+ I >>is 221. 1 ±26. 1 ( P < 0. 01) . Conclusion  These data indicate that verapamil can stop the influx of Ca2+ and hence inhibit the FCS 3/ induced proliferation signal t ransduction.
