  • 摘要: 靶向近红外荧光(Near-Infrared Fluorescence, NIRF)探针因其在肿瘤早期诊断、术中导航和个体化治疗中的独特优势,受到广泛关注。近红外荧光具有较强的组织穿透性、低背景干扰和光损伤小等特点,使其成为理想的体内分子成像工具。通过将NIRF染料与高亲和力的靶向基团结合,靶向探针能够特异性识别肿瘤表面标志物(如FR、PSMA、HER2、EGFR、CEA等),增强肿瘤区域的荧光信号和对比度。随着荧光染料的优化和靶向基团的升级,靶向NIRF探针在多个恶性肿瘤中的应用持续拓展,尤其在肿瘤术中成像、影像检查和治疗中展现了良好的前景。本文综述了靶向NIRF探针的研究进展,重点讨论了其结构改良、临床前研究和初步临床应用探索。

     

    Abstract: Targeted near-infrared fluorescence (NIRF) probes have garnered widespread attention due to their unique advantages in early tumor diagnosis, intraoperative navigation, and personalized therapy. NIR fluorescence offers strong tissue penetration, low background interference, and minimal phototoxicity, making it an ideal tool for in vivo molecular imaging. By conjugating NIRF dyes with high-affinity targeting groups, these probes can specifically recognize tumor-associated biomarkers (such as FR, PSMA, HER2, EGFR, CEA, etc.), enhancing fluorescence signal intensity and contrast in tumor regions. With the continuous optimization of fluorescent dyes and upgrading of targeting groups, targeted NIRF probes are expanding their applications in various malignant tumors, showing great potential in tumor intraoperative imaging, diagnostic imaging, and therapy. This review summarizes recent advances in targeted NIRF probes, focusing on structural improvements, preclinical studies, and early clinical applications.
