基于双样本孟德尔随机化探讨结直肠癌与常见精神类疾病的因果关系

Colorectal Cancer and Common Psychiatric Disorders: A Two-sample Mendelian Randomization Study

  • 摘要: 目的:基于双样本孟德尔随机化(2-Sample Mendelian randomization, 2-Sample MR)分析结直肠癌与常见精神类疾病的因果关系。方法:依托于公共数据库中结直肠癌与精神类疾病的GWASs数据,通过逆方差加权、MR-Egger和中位数加权三种方法评估结直肠癌与抑郁症、焦虑症、双向情感障碍和精神分裂症之间的关系,最后通过敏感性分析评估结论的可信度和稳健性。结果:逆方差加权显示结直肠癌与抑郁症(OR=0.995,95%CI:0.995~1.000,P=0.090)、焦虑症(OR=1.001,95%CI:1.000~1.001,P=0.099)、精神分裂症(OR=1.003, 95%CI:0.9603~1.0473,P=0.899)之间没有因果关系;但与双向情感障碍风险降低存在因果关系(OR=0.937,95%CI:0.8919~0.9849,P=0.010)。结论:研究表明,结直肠癌与抑郁症、焦虑症和精神分裂症无因果关系,但结直肠癌与双向情感障碍风险降低可能存在因果关联,同时印证了肠脑轴的存在。

      Objective: This study aimed to elucidate the causal relationships between colorectal cancer (CRC) and prevalent psychiatric disorders using a two-sample Mendelian randomization approach, and provide evidence supporting the gut-brain axis (GBA) hypothesis.

      Methods: Utilizing publicly available GWAS datasets, we explored the connections between CRC and various psychiatric disorders, including depression, anxiety, bipolar disorder, and schizophrenia. We applied three statistical analyses: inverse variance weighting, MR-Egger, and median weighting. Sensitivity analyses were conducted to ensure the reliability and validity of our results.
