Cyclin F Expression in Clear Cell Renal Cell Carcinoma and Its Effect on Biological Behavior of Renal Carcinoma Cell Lines

    目的 探讨细胞周期蛋白F(Cyclin F)在肾透明细胞癌(ccRCC)中的表达、临床病理特征及其对肾癌细胞株生物学行为的影响。
    方法 RT-qPCR和Western blot分别检测新鲜ccRCC标本中Cyclin F mRNA和蛋白表达。免疫组织化学法检测80例石蜡包埋样本中Cyclin F蛋白表达;CCK-8实验、划痕实验、流式细胞术检测过表达Cyclin F对肾癌细胞增殖、迁移和凋亡的影响。
    结果 Cyclin F mRNA在癌组织中表达高于癌旁组织(P<0.0285),Cyclin F蛋白在癌组织中表达低于癌旁组织(P<0.001),临床病理参数分析显示,Cyclin F低表达与患者WHO/ISUP分级、Ki67指数和年龄显著相关(P=0.041,0.047,0.008,)。体外实验证实,过表达Cyclin F促进肾癌786-O细胞株的增殖(P<0.001)和迁移能力(P24 h=0.003, P48 h=0.0058),抑制细胞凋亡(P=0.0019)。
    结论 Cyclin F在ccRCC中蛋白低表达与患者高ISUP分级、高Ki67指数、高年龄及预后有关。

     

    Abstract:
    Objective To investigate the expression of Cyclin F in clear cell renal cell carcinoma (ccRCC), its clinicopathological characteristics, and its effect on the biological behavior of renal cancer cell lines
    Methods RT-qPCR and Western blot were used to detect the mRNA and protein expression of Cyclin F in fresh ccRCC specimens. Immunohistochemistry assay was performed to detect the expression of Cyclin F protein in 80 paraffin samples. CCK-8 assay, scratch assay, and flow cytometry were conducted to determine the effects of Cyclin F overexpression on the proliferation, migration, and apoptosis of renal cancer cell lines.
    Results The expression of Cyclin F in cancer tissues was higher than that in adjacent tissues at the mRNA level (P<0.0285). The expression of Cyclin Fprotein in cancer tissues was lower than that in adjacent tissues (P<0.001). The analysis of clinicopathological parameters showed that the low expression of Cyclin F was correlated with WHO/ISUP grade, Ki67 index, and age (P=0.041, 0.047, 0.008,). In vitro experiments confirmed that overexpression of Cyclin F promoted the proliferation (P<0.001) and migratory ability (24 h P=0.003, 48 h P=0.0058) of the RCC 786-O cell line, while inhibiting apoptosis (P=0.0019).
    Conclusion The low expression of Cyclin F protein in ccRCC tissuesis associated with high ISUP grade, high Ki67 index, oldage, and prognosis of patients.
