宫颈癌组织中OPNHIF-1α的表达及其与新辅助化疗敏感度的关系

Relationship between Expressions of OPN and HIF-1α and Neoadjuvant Chemotherapy Sensitivity in Cervical Carcinoma Tissues

  • 摘要: 目的 探讨新辅助化疗前、后宫颈癌组织OPN mRNA与HIF-1α mRNA的表达变化,分析其作为宫颈癌新辅助化疗敏感度预测因子的可能性。方法 采用实时荧光定量PCR法检测20例宫颈癌患者新辅助化疗前、后组织中OPN mRNA与HIF-1α mRNA的表达水平。结果 宫颈癌患者新辅助化疗后组织中OPN mRNA与HIF-1α mRNA的表达水平明显下降(P<0.05)。新辅助化疗有效组化疗前、后组织中OPN mRNA与HIF-1α mRNA表达水平均明显低于无效组(P=0.016,P=0.001)。结论 宫颈癌患者组织中OPN mRNA与HIF-1α mRNA的表达水平与化疗的敏感性及临床疗效关系密切,可作为宫颈癌新辅助化疗敏感度的重要预测因子。

     

    Abstract: Objective To investigate the mRNA expression and significance of OPN and HIF-1α pre- and post-neoadjuvant chemotherapy in cervical carcinoma, and analyze its possibility to be the predictors for neoadjuvant chemotherapy sensitivity. Methods Real-time quantitative reverse transcription-polymerase chain reaction(qRT-PCR) method was used to detect the mRNA levels of OPN and HIF-1α in pre- and postneoadjuvant chemotherapy in 20 cases of cervical carcinoma tissues. Results The mRNA levels of OPN and HIF-1α were signifi cantly decreased after neoadjuvant chemotherapy in patients with cervical cancer tissues (P<0.05). mRNA levels of OPN and HIF-1α in effective chemotherapy group were lower than ineffective group pre- and post-neoadjuvant chemotherapy(P=0.016, P=0.001). Conclusion OPN and HIF-1α mRNA levels were signifi cantly associated with neoadjuvant chemotherapy chemosensitivity and they might be used as predictors for neoadjuvant chemotherapy sensitivity in cervical carcinoma.
