Effect of RetroNectin on Proliferation and Biological Characteristics of CIK Cells in Hepatocellular Carcinoma (HCC) Patients

  • 摘要: 目的 观察重组人纤维连接蛋白(RN)对肝细胞肝癌(HCC)患者细胞因子诱导的杀伤细胞(CIK)生物活性的影响,建立一种高效的CIK细胞体外扩增方法。方法 取15份HCC患者外周血,分离单个核细胞,分别使用传统方法(抗CD3 mAb+IFN-γ+IL-2)和RN法(RN+抗CD3 mAb+IL -2)诱导,观察不同时间点两组细胞形态、数目、细胞表型和杀伤效率。结果 与传统方法相比,RN组显著增加CIK细胞扩增倍数(P<0.05),是对照组的2.0~2.8倍,提高活化T细胞(CD25+T细胞)数量,增加CD3+CD8+T细胞比例,增强对肝癌细胞株的杀瘤活性。结论 RN能够高效促进HCC患者CIK细胞的扩增,可以替代传统方法。

     

    Abstract: Objective To observe the biological characteristics of CIK cells activated by recombinant human fibronectin(RN) and establish an effective way of cell proliferation. Methods We separated the mononuclear cells(PBMCs) from 50 ml peripheral blood from 15 HCC patients PBMCs were divided into two groups,of which were activated by RN-induced (RN+anti-CD3 mAb+IL-2) or conventional method(anti-CD3 mAb+IFN-γ+IL-2) repectively.Then we estimated cell morphology,the number,cell phenotype and cytotoxic efficiency at different time points in two groups. Results RN-induced group had a 2.0~2.8 fold higher proliferation rate,compared with conventional group,P<0.05.There was also a higher percentage of activated T cells and CD3+CD8+T cells in RN-induced group.We observed a higher inhibition rate of the CIK cells to liver cancer cell lines. Conclusion RN may replace conventional method to induce CIK cell proliferation in HCC patients.
