  • 摘要: 目的 探讨骨桥蛋白(osteopontin,OPN)在ⅠA~ⅡA期宫颈浸润癌中的表达及其与宫颈癌病理特征的关系和在宫颈癌淋巴结转移中的作用。方法 运用链霉菌抗生物素蛋白-过氧化物酶法(SP法)检测44例ⅠA~ⅡA期宫颈浸润癌患者及15例正常宫颈组织的骨桥蛋白水平。结果 (1) 44例ⅠA~ⅡA期宫颈癌组织中骨桥蛋白的阳性表达率为72.73%(32/44),高于正常宫颈组织的13.33%(2/15),两组比较差异有统计学意义(P<0.05)。(2) 骨桥蛋白的表达在宫颈癌分化程度、组织类型、浸润深度、患者年龄间比较,差异均无统计学意义(P>0.05)。而在ⅠA、ⅠB期与ⅡA期,在小于或等于1个淋巴结转移患者与大于1个淋巴结转移之间患者比较,差异有统计学意义(P<0.05)。结论 骨桥蛋白在宫颈癌中呈阳性表达,其阳性表达与临床分期、淋巴结转移数目有关,检测宫颈癌组织骨桥蛋白表达可为预测盆腔淋巴结转移提供帮助。

     

    Abstract: Objective To study the effect of rotary magnetic field (RMF) combining 5-Fu on the cycle and apoptosis of mouse cell line SP2/0 in vitro. Methods SP2/0 cells were randomly divided into four groups: control group (N), 5-Fu group (C), magnetic group (M) and magnetic combining 5-Fu group (M+C).The M and M+C groups were treated with a RMF for two hours once a day.On day 4, the C and M+C groups were treated with 5-Fu 20 μg/ml.On day 5, cell cycle and apoptosis were measured by the flow cytometric (FCM). Results The S phase proportion of the M group and the G1 phase proportion of the C group were higher than that of the other three groups(P<0.05).The S phase proportion of the M+C group decreased and lower than that of the M group,but was still higher than that of the N and C groups(P<0.05).There was no significant difference in apoptosis rates between the N and M groups(P>0.05).The apoptosis rates of the C and M+C groups were remarkedly higher than those of the N and M groups and the M+C group had the highest apoptosis rate. Conclusion The RMF can't induce the apoptosis.But it can enhance the cytotoxicity of 5-Fu and promote the cell apoptosis.The mechanism of the apoptosis may be related to SP2/0 cell line arrested at S phase.Objective To investigate the expression levels of osteopontin (OPN) in stage ⅠA~ⅡA cervical carcinoma and to study the pathological characteristics.To discuss their contribution to lympho-metastasis. Methods The expression of OPN was detected in 44 samples of cervical carcinoma with stage ⅠA~ⅡA and 15 samples of normal cervix by using SP method (streptavidin-perosidase,SP) to analyze the correlation with clinical pathological characters, lympho-metastasis and their clinical implication.Results (1)The positive expression rates of OPN was 72.73%(32/44) and 13.33% (2/15) in cervical carcinoma and normal cervix respectively with significant difference(P<0.05).(2) The positive expression rates of OPN had no significant in cervical cancer's pathological grades, tissue types, invasive depth and ages(P>0.05).But the expression had significant differences in stage Ⅰa, Ⅰb and stage Ⅱa(P<0.05).OPN expression in patients with less than or equal one positive lymphonode was different from others with more than one positive lymphonodes(P<0.05). Conclusion The positive expression rates of OPN in cervical carcinoma were higher and has significant differences in clinical stages.The positive expression of OPN corelates to amount of positive lymph nodes. The expression of OPN may be helpful to predicte lympho-metstasis.
